The second installment of Hataraku Oniisan! continues the story of lively Calico Tapio Chatorazawa and perpetually grumpy Russian Blue Kuehiko Roshihara as they take on a range of part-time jobs together. Being classmates, they experience both the difficulties and rewards that come with their work endeavors.

Tapio Chatarozawa, full of energy, and the composed Kuehiko Roshihara are two young felines who take on a variety of part-time jobs. From delivering parcels to washing windows on skyscrapers, the duo plunges into the working world, uncovering new opportunities at every turn. Despite the difficulties and the strange habits of their customers and supervisors, Tapio and Kuehiko forge a close friendship as they manage their many different work experiences.

After making it through the attack at Sarugaku headquarters, Sousuke Banba continues his quest to track down the Kagewani monster hybrids. He now possesses the ability to tap into the Kagewani's own power, giving him a weapon to destroy these creatures. However, this skill comes with a serious cost: Sousuke is fighting to hold onto his sanity as the power slowly eats away at his body. New and familiar dangers soon emerge—the higher ranks of Sarugaku become active again, and a female assassin driven by a personal grudge targets Sousuke. Still, he presses on without hesitation, determined to uncover the source of the Kagewani and put an end to the threat for good.

A video blogger fakes encounters with mythical beasts to boost his channel's ratings, but soon faces a real monster that annihilates his crew. Meanwhile, students are attacked by a sandworm-like creature, forcing them into a desperate struggle for survival within their school. As a wave of mysterious assaults escalates, researcher Sousuke Banba sets out to solve the mystery, guided only by the cryptic word "Kagewani." He examines the locations of the latest incidents, seeking a lasting way to eliminate the threats. Yet his investigation runs into trouble when the pharmaceutical company Sarugaku begins blocking his moves, hinting that the peril to humanity may be far nearer than he imagined.
