In the metropolis of Stern Bild, individuals with special powers are known as NEXT, using their abilities for either heroic or criminal purposes. A select group of these NEXT appear on Hero TV, pursuing villains to earn points for their sponsors and compete for the title of King of Heroes. Kotetsu T. Kaburagi, also called "Wild Tiger," is a veteran hero whose performance has recently declined, partly due to his difficulty working with others. After a lackluster season where his colleagues far outperformed him, Tiger is paired with a new partner, Barnaby Brooks Jr., who goes by his real name as his hero alias. Nicknamed "Bunny" by his laid-back partner, Barnaby quickly proves to be very different from Tiger. In spite of their opposing natures, Tiger and Bunny must learn to cooperate, both for the sake of their careers and to face the growing dangers in their city.

Soon after, Goemon Ishikawa XIII, Fujiko Mine, and Kouichi Zenigata all get pulled into the situation, each for their own reasons. As everyone ends up at Cagliostro Castle, Lupin remembers a visit he made there ten years earlier, and the hidden truths about the castle begin to come out.

In this installment, Lupin takes center stage against the backdrop of Italy and San Marino. Following its geographical theme, the anime made its debut in Italy prior to airing in Japan. Fans of Lupin III will notice a change in his attire, as he now sports a blue jacket, differing from his previous ensembles including green (from the first series and certain OVAs), red (featured in the second series, most films, and TV specials), and pink (from the third series).

A wealthy local entrepreneur intends to recover this gem, and once it goes on display at the Marina Sands hotel in Singapore, a murder occurs, accompanied by a bloody calling card from Kaitou Kid. Concurrently, Ran and Sonoko travel to Singapore for a karate competition. Conan, unable to leave Japan because he lacks a passport, is supposed to stay behind. However, Kaitou Kid, in need of Conan's aid, uses a magic trick to forcibly bring Conan to Singapore. To return home, Conan must cooperate with Kid, but first he has to disguise himself since his glasses, watch, and other belongings are taken away. When Ran fails to identify him, he calls himself "Arthur Hirai." Later, Kid discovers that the "fist" is stored in a concealed vault beneath a mansion. As he thinks he has safely accessed it, a dangerous trap is set off. He then confronts Makoto Kyogoku, the undefeated karate champion boasting 400 consecutive victories.

A mystery emerges during the Justice Day celebration in Sternbild, an event that honors the city's revered goddess, and drives the film's plot.

The Danger Service Agency (DSA) is composed of three members: Mikura, Kurokawa, and Harada. Mikura provides the physical brawn, Harada supplies the brains, and Kurokawa is a bitter ex-cop who likes to believe he is in charge. They accept any assignment that involves plenty of risk and, of course, payment. Crossing them is unwise if you want to live long enough to eat dinner. The most significant case they face may be uncovering why someone wants Kurokawa assassinated.
